Design and construction of the CasRx-crRNA-Ribozyme system (CCRS). a Design of the fusion guide RNA construct of CCRS. The CasRx guide RNA has a 5′-scaffold sequence in front of the antisense RNA sequence, and the ribozyme shares the same 5′-antisense RNA with crRNA. The fusion guide RNA has scaffolds for CasRx and ribozyme, enabling RNA cleavage with both CasRx and ribozyme. H at position 7 designates A, C, or U.
