Core meiotic recombination components are required for normal meiosis in Sd. ludwigii. a Meiotic time-course analysis of a homozygous Δspo11 Sd. ludwigii mutant in comparison to its wild type (left). After induction of meiosis, samples were withdrawn at the indicated time points and their cellular DNA content was stained with Hoechst 33258 to determine the fractions of binucleate (meiosis I) and tetranucleate (meiosis II) cells. Error bars: SD (n = 3 replicates for each strain). Spore viability was examined using tetrad dissection and viable colony counting (right). A total of 50 tetrads of each strain were analyzed. b–d Meiotic nuclear spreads of sporulating cells with the indicated genotypes (homozygous diploids). Cellular DNA was stained with Hoechst 33258.
