Analysis of the rs4919742-associated chromatin loops. Guide RNAs targeting regions encompassing CTCF site 4 (the PCa risk-associated CTCF site), CTCF site 5, and/or CTCF site 6 (or the empty guide RNA vector as a control) were introduced into 22Rv1 prostate cancer cells, along with Cas9. Cell pools were harvested, and KRT78 expression was analyzed by RT-qPCR. Shown within the blue bars is the fold change in KRT78 expression in the pools that received guide RNAs vs the vector control.
