Differential gene expression during planarian regeneration. (a) For the longest isoform of each locus from the Illumina+ assembly, the expression fold change (log2 scale) of the 2 to 3 h sample relative to the control (0 h) is plotted against its log average abundance (MA plot). Statistically significant up- or down-regulation (adjusted P-value < 0.001 and log2 fold change > 0.7 or < -0.7 (red lines)) is indicated in yellow and blue, respectively. Genes chosen for quantitative RT-PCR (qRT-PCR) validation are labeled. (b) Comparison of results from the Illumina transcriptome sequencing (RNAseq) time course with an independent head and tail amputation experiment assayed by qRT-PCR. Expression levels were normalized to that of intact controls (time point 0). RNAseq results are shown at the mid-point of the respective window of time covered by samples in each library. Error bars for qRT-PCR data represent standard errors of the mean of three biological replicates.
