BAX has four motifs: BH3 motif (PS01259), BH1 (PS01080), BH2 (PS01258), and BCL2-like apoptosis inhibitor family profile (PS50062). BH3 (in red) has the highest change in log-likelihood among those motifs, and is among one of our top predictions (1.9%). Reed et al. [48] confirmed that BH3 on BAX is involved in binding with BCL2. On the other hand, the binding site on BCL2 involves portions where all of BH1, BH2, and BH3 reside. Interestingly, none of these motifs on BCL2L1 have high confidence to be a binding site, with the highest one also being BH3 and ranked in the top 8.7%. Mutations in BAX (in position shown by the black bar) cause leukemia.
