(a) Schematic representation of the purification protocols used to purify the HMT-histone complexes and crude histones. (b) Doubly immunopurified Suv39h1 complexes from chromatin extracts of 20 g of HeLa-Suv39h1 cells were resolved on a 4-12% gradient NuPAGE gel, run in MES buffer (Invitrogen), fixed, and stained with Colloidal blue. Lane 1, SeeBlue pre-stained molecular weight markers (Invitrogen); lane 2, Suv39h1 complex from chromatin fractions.
