The relationship between cohesin and its loader. (a) Deletion of two tRNA genes removes a cohesin loading site, but does not affect the surrounding cohesin pattern. Mis4-Pk9 and Rad21-Pk9 association over a 35 kb region of chromosome 2 is compared between strains containing, or deleted for, the two tRNA genes SPBTRNAARG.04 and SPBTRNAGLY.05. Grey bars above and below the midline represent ORFs transcribed from left to right and opposite, respectively. The two tRNA genes are highlighted in red. (b) Distance distribution of cohesin-bound and cohesin-free convergent sites to their nearest Mis4 binding sites. Boxes indicate boundaries of the 25th to 75th percentile surrounding the median (solid line). Whiskers encompass 1.5 times the interquartile range, outliers are indicated. A Wilcoxon signed ranks test suggests that cohesin-bound convergent sites are closer to their nearest loading site than cohesin-free convergent sites.
